    Obiettivi formativi

    Il corso si pone come obiettivo quello di fornire allo studente le conoscenze riguardanti lo studio della struttura, funzione e regolazione delle macromolecole biologiche, quali il DNA, l’RNA e le proteine focalizzando l'attenzione sulle vie biochimiche in cui sono coinvolte. In particolare, verranno affrontati i meccanismi alla base della replicazione e riparazione del DNA e quelli che controllano il flusso dell'informazione genetica dal DNA alle proteine, trascrizione e traduzione, e i relativi meccanismi di regolazione cellulare. In questo modo lo studente sarà in grado di comprendere e conoscere la struttura, la funzione e l'attività delle macromolecole biologiche, e delle vie coinvolte nella regolazione e controllo del flusso dell’informazione biologica.

    Prerequisiti

    Non esiste alcuna propedeuticità sebbene è necessario aver acquisito conoscenze di base di chimica generale.

    Metodologie didattiche

    Il corso è organizzato in lezioni frontali con supporto informatico (Power Point) e cartaceo (documenti, lavori scientifici in pdf).

    Metodi di valutazione

    La verifica dell'apprendimento avverrà tramite colloquio orale. Le domande riguarderanno gli aspetti di base e principali della biologia molecolare. Pertanto, i quesiti risulteranno specifici per valutare le conoscenze fondamentali dell’argomento. Lo studente dovrà dimostrare di conoscere gli aspetti chiave della struttura, della funzione e della regolazione delle macromolecole biologiche, come il DNA, l'RNA e le proteine. Verranno analizzate le capacità dello studente di discutere su questi punti e l’abilità nello stabilire possibili connessioni. Il voto si esprima in trentesimi, partendo da un minimo di 18 fino ad un massimo di 30/30 e lode.

    Acidi Nucleici.
    Struttura e proprietà dei nucleotidi. I nucleotidi sono formati da basi, zuccheri e fosfati caratteristici. I legami fosfodiesterici uniscono le unità nucleotidiche negli acidi nucleici. Le basi nucleotidiche influenzano la struttura tridimensionale degli acidi nucleici. Altri ruoli dei nucleotidi nelle cellule.
    Struttura del DNA
    Le molecole di DNA hanno una tipica composizione in basi. Solitamente il DNA è una doppia elica destrorsa. Il DNA adotta diverse forme di elica. Caratteristiche fisiche della molecola di DNA : denaturazione e rinaturazione.
    Struttura dell’RNA
    Le molecole di RNA hanno una tipica composizione in basi. RNA World: mRNA, tRNA, rRNA, piccoli RNA ed altri RNA.
    La replicazione del DNA.
    Esperimento di Meselson e Stahl. Caratteristiche generali della duplicazione del DNA. Replicazione nei batteri. La struttura e il ruolo biologico delle differenti DNA polimerasi procariotiche. Attività esonucleasica della DNA polimerasi III: la correzione delle bozze. La replicazione negli eucarioti. La struttura e il ruolo biologico delle differenti DNA polimerasi eucariotiche. Antigene nucleare di proliferazione cellulare (PCNA) collegamento tra duplicazione e ciclo cellulare. Telomeri e telomerasi: il problema delle estremità nel genoma eucariotico.
    Mutazioni e meccanismi di riparo del DNA.
    Tipi di mutazioni. Mutazioni spontanee: errori della duplicazione, tautomeria delle basi, espansioni di triplette. Mutazioni indotte: agenti chimici, agenti fisici. Agenti chimici: Analoghi di basi, agenti deaminanti, agenti alchilanti e agenti intercalanti. Agenti fisici: radiazioni UV, radiazioni ionizzanti, il calore. Sistemi di Riparo: riparazione diretta, riparazione per escissione di basi e di nucleotidi, riparazione dei mismatch, riparazione per ricombinazione. Meccanismo SOS.
    La trascrizione dell’RNA.
    Caratteristiche generali della trascrizione. La struttura del promotore procariotico. Trascrizione nei batteri. RNA polimerasi procariotica e l’importanza dei fattori sigma. Terminatori intrinseci e la proteina rho. Maturazione degli RNA nei batteri. La trascrizione negli eucarioti.     Proteine
    Le tappe della biosintesi delle proteine: il codice genetico e le sue caratteristiche, ruolo di tRNA, aminoacil-tRNA sintetasi, fattori di inizio, elongazione e rilascio in procarioti ed eucarioti. Inibitori della sintesi proteica.
    Modifiche post-traduzionali (ripiegamento proteico, taglio proteolitico, modificazione chimica e splicing delle inteine), smistamento e degradazione delle proteine (il sistema ubiquitina-proteosoma).
    Regolazione dell’espressione genica
    Regolazione dell’espressione genica in procarioti: l’esempio degli operoni lattosio e triptofano.
    Regolazione dell’espressione genica negli eucarioti: l’esempio dell’RNA antisenso ed il meccanismo dell’interferenza da RNA (siRNA).

    Course objectives

    The course aims to provide students with the knowledge concerning the study of the structure, function and regulation of biological macromolecules, such as DNA, RNA and proteins, focusing attention on the biochemical pathways in which they are involved. In particular, the mechanisms underlying DNA replication and repair will be addressed, as well as those that control the flow of genetic information from DNA to proteins, transcription and translation, and the related mechanisms of cell regulation. In this way the student will be able to understand the structure, function and activity of biological macromolecules, and of the routes involved in the regulation and control of the flow of biological information.

    Prerequisites

    There are no prerequisites, although it is necessary to have acquired basic knowledge of general chemistry.

    Teaching methods

    The course is organized in lectures with computer support (Power Point) and paper (documents, scientific works in pdf).

    Evaluation methods

    Evaluation of student proficiency is based on and oral test (viva voce). Questions cover the basic aspects as well as the main aspects of molecular biology. The questions will be as specific to evaluate the basic knowledge of the topic. This means that the student must know the key steps of the structure, function and regulation of biological macromolecules, such as DNA, RNA and proteins. The student should be able to discuss on these topics and be able to connect and analyze the different subjects. The final grade is expressed in 30/30 were 18 represents the minimum and 30 the maximum.

    Nucleic Acids.
    Structure and properties of nucleotides the nucleotides are made of bases, sugars and phosphates characteristic. Phosphodiester bond combine nucleotide units in nucleic acids. The nucleotide bases influence the three-dimensional structure of nucleic acids. Other roles of nucleotides in cells.
    DNA structure.
    DNA molecules have a typical base composition. Usually DNA is a double helix right-handed. The DNA helix shows different shapes. Physical characteristics of the DNA molecule: denaturation and renaturation.
    RNA structure
    RNA molecules have a typical base composition. RNA World: mRNA, tRNA, rRNA, small RNA and other RNA.
    DNA replication.
    Experiment by Meselson and Stahl. General features of DNA duplication. Replication in bacteria. The structure and the biological role of the different prokaryotic DNA polymerases. Exonuclease activity of DNA polymerase III: proofreading. Replication in eukaryotes. The structure and the biological role of the different eukaryotic DNA polymerases. Proliferating cell nuclear antigen (PCNA) link between duplication and cell cycle. Telomere and telomerase: the problem of extremities in the eukaryotic genome.
    Mutations and mechanisms of DNA repair.
    Types of mutations. Spontaneous mutations: duplication errors, Keto–enol tautomerism, Trinucleotide Repeat Expansion. Induced mutations: chemical agents, physical agents. Chemical agents: base analogues, deaminating agents, alkylating agents and intercalating agents. Physical agents: UV radiation, ionizing radiation, heat. Repair Systems: direct repair, nucleotide excision repair and base excision repair, mismatch repair, recombination repair. SOS mechanism.
    RNA transcription.
    General characteristics of the transcription. The structure of the prokaryotic promoter. Transcription in bacteria. Structural Biology of Bacterial RNA Polymerase and the key rule of sigma factors. Intrinsic terminators and the rho protein. RNA maturation in bacteria. Transcription in eukaryotes. Characteristics and structure of eukaryotic RNA polymerases: comparison between eukaryotic promoters. The transcription pre-initiation complex (PIC) and the role of general transcription factors in eukaryotes.
    Maturation and modifications of the messenger RNA: signal sequences, capping, splicing and alternative splicing, self-splicing and polyadenylation. Editing

    Protein
    The stages of protein biosynthesis: the genetic code and its characteristics, role of tRNA, aminoacyl-tRNA synthetase, start factors, elongation and release in prokaryotes and eukaryotes. Post-translational modifications (protein folding, proteolytic cutting, chemical modification and splicing of inteins), sorting and degradation of proteins (the ubiquitin-proteasome system).
